PEPPER lance la validation de deux nousvelles méthodes « Test de Transactivation du Récepteur Activé par les Proliférateurs de Peroxysomes γ (PPARγ-TA) », « Test de Transactivation du Récepteur de la Progestérone (PR-TA) »

Résumé des deux méthodes :

« Test de Transactivation du Récepteur Activé par les Proliférateurs de Peroxysomes γ (PPARγ-TA) » : PPARγ est un récepteur clé impliqué dans la différenciation des adipocytes, les réponses immunitaires, la prolifération tissulaire et le métabolisme, notamment le stockage des graisses. Cible de longue date pour la découverte de médicaments, il est notamment visé par les glitazones, qui améliorent la sensibilité à l’insuline. De nombreux composés peuvent interagir avec PPARγ, mais l’hétérodimérisation avec le récepteur RXR rend difficile la distinction entre leurs effets respectifs. La signalisation PPARγ est aussi liée au développement de certaines tumeurs. Un test in vitro basé sur l’activation d’un gène rapporteur de la luciférase permet de mesurer l’activation ou l’inhibition de PPARγ humain par des substances chimiques. Ce test peut également être utilisé en mode antagoniste, avec la rosiglitazone comme agoniste de référence.
« Test de Transactivation du Récepteur de la Progestérone (PR-TA) » : La progestérone est importante pour divers processus et fonctions de l’organisme, notamment la grossesse, la neuroprotection, le développement comportemental, la formation des os et le métabolisme. Souvent, elle agit en collaboration avec d’autres hormones. En outre, la perturbation de la signalisation du récepteur de la progestérone (PR) est associée à des résultats négatifs tels qu’un risque accru de développer des cancers hormonodépendants. Cette méthode in vitro mesure le potentiel des substances chimiques à activer ou à inhiber le PR par la régulation d’un gène rapporteur de la luciférase. Comme d’autres tests de transactivation, le PR-TA repose sur la liaison et l’activation de substances chimiques au PR pour induire la luciférase. En outre, la méthode
peut être transformée en un essai antagoniste utilisant la progestérone qui peut être concurrencée par les produits chimiques testés.
